Advantages and Limitations

Furthermore, it allows for the assessment of RNA quality, as degradation patterns are easily visible on the gel.

However, the technique has notable limitations. Additionally, it is more labor-intensive and time-consuming, and it involves the use of specialized probes and, historically, radioactive labels, necessitating careful handling and disposal protocols.

While high-throughput sequencing technologies have largely superseded Northern blotting in large-scale transcriptomic studies, the Northern blot remains a gold standard for validating the results of other gene expression assays. It is frequently employed in research to confirm findings from microarray or RNA-Seq experiments, particularly when characterization of transcript size is of specific interest.
